Acclimatization of micropropagated Heliconia bihai (Heliconiaceae) plants Scientia Agricola

To evaluate the behavior of micropropagated heliconia seedlings during the acclimatization process using different substrates and shade levels, seedlings of H. bihai (Lobster Claw I) were planted in plastic trays using the substrates: washed sand, vermiculite (medium texture), and PlantMax® Horticultura, and shade cloths with percentages of 0% (full sun), 30%, 40%, 50%, 60%, 70%, and 80% of shade in relation to full sun. Best results were obtained for washed sand and PlantMax substrates at shade conditions starting from 50%, especially at 70% and 80%. Pre-existing roots from in vitro rootings were not functional, not presenting secondary roots, and showing root tissue necrosis. Acclimatized seedlings developed new roots. Agrobacterium rhizogenes vs auxinic induction for in vitro rhizogenesis of Prosopis chilensis and Nothofagus alpina Biocell

The induction and improvement of in vitro rhizogenesis of microshoots of Prosopis chilensis (Mol.) Stuntz and Nothofagus alpina (Poep. et Endl. Oerst.) were compared using Agrobacterium rhizogenes (Ar) versus indole-3-butyric acid (IBA) in the culture media. Microshoots of P. chilensis (1-2 cm length), coming from in vitro grown seedlings, were cultivated in a modified Broadleaved Tree Medium (BTMm) containing half salt concentration of macronutrients and 0.05 mg.L-1 benzilaminopurine (BAP). Arbuscular mycorrhizal fungi and auxin associated with microelements in the development of cuttings of Varronia leucocephala AGRIAMBI

ABSTRACT The plant Varronia leucocephala is widely used in Brazil for its therapeutic properties. However, a major problem for the seedlings is the low percentage of root formation. The objective of this study was to establish a rooting protocol for V. leucocephala cuttings, using phytoregulators and microelements associated with arbuscular mycorrhizal fungi. The auxin indole-3-butyric acid (IBA) concentration of 1500 mg L-1 showed the best rooting percentage, and it is proposed associating the microelements zinc and boron with the highest IBA doses. ABSTRACT This study aimed to evaluate the potential of multi-stems in avocado seeds according to their mass as well as the adventitious rooting of multi-stem budding with or without the use of auxin. Comparison of Root Induction in Mature Filbert (Corylus avellana L.) Explants by Agrobacterium Rhizogenes and Indolbutiric Acid Chilean J. Agric. Res.

From in vitro cultured adult material of Corylus avellana L. cv. Negretta, adventitious rooting of microshoots was evaluated. Rhizogenic induction mediated by two strains of wild-type Agrobacterium rhizogenes (A477 and A478) and indolbutyric acid (IBA) were compared under two light conditions (16:8 h photoperiod and complete darkness). The results indicate that in the 16:8 h photoperiod induction, the rooting rate with IBA (90%) was significantly higher than that obtained with the strain A477 of A. rhizogenes (67.7%), while with the strain A478 no statistically significant difference was obtained for the same variable (75%). Early selection of Cabralea canjerana for propagation by mini-cutting PAB

Abstract: The objective of this work was to define an early selection strategy to identify Cabralea canjerana (Meliaceae) clones with high multiplication rate. A mini-garden of 109 clones of canjerana seedlings was established in a completely randomized design, in an acclimatized greenhouse. From seedlings, the mini-stumps and mini-cuttings were obtained. Mini-cuttings were collected at five different times, and the number of mini-cuttings per mini-stump, rooting percentage, and number of rooted mini-cuttings were quantified. The number of rooted mini-cuttings per mini-stump was the only trait that showed high correlation with the others. Este trabalho teve como objetivo verificar o efeito de seis concentrações de ácido indolbutírico (IBA) no enraizamento de estacas herbáceas de figueira (Ficus carica L.) sob nebulização intermitente, em três épocas diferentes. Os ensaios foram instalados em maio/2004, setembro/2004 e janeiro/2005, e, em cada época, o delineamento experimental utilizado foi o inteiramente casualizado, com seis concentrações de IBA (0, 500, 1.000, 1.500, 2.000, 2.500 e 3.000 mg L-1). Para avaliar o efeito de cinco concentrações de IBA utilizou-se os substratos areia lavada e cinza de casca de arroz, no enraizamento de estacas cilíndricas e triangulares de laranjeira cv. valência, coletadas em três épocas do ano. Os resultados mostraram que os tratamentos não influenciaram a formação de raízes; a areia foi o melhor substrato para formação de calos sendo a melhor concentração, 4500ppm de IBA para estacas cilíndricas coletadas em maio.

Effects of plant growth promoting rhizobacteria (PGPR) on rooting and root growth of kiwifruit (Actinidia deliciosa) stem cuttings Biol. Res.

The effects of plant growth promoting rhizobacteria (PGPR) on the rooting and root growth of semi-hardwood and hardwood kiwifruit stem cuttings were investigated. The PGPR used were Bacillus RC23, Paenibacillus polymyxa RC05, Bacillus subtilis OSU142, Bacillus RC03, Comamonas acidovorans RC41, Bacillus megaterium RC01 and Bacillus simplex RC19. All the bacteria showed indole-3-acetic acid (IAA) producing capacity. Among the PGPR used, the highest rooting ratios were obtained at 47.50% for semi-hardwood stem cuttings from Bacillus RC03 and Bacillus simplex RC19 treatments and 42.50% for hardwood stem cuttings from Bacillus RC03. ABSTRACT In order to establish an optimal soybean embryonic tip regeneration system that can serve as soybean genetic transformation receptor, and be used for the study of genetic function verification, the influences of single factor on the adventitious bud of embryonic tip induction, elongation and rooting stage, are researched and compared.The single factors includes seeds soaking time, different kinds of hormones, different concentration of hormone and different concentration of sucrose. Starting from semihardwood branches of peach and plum rootstocks (Prunus cerasifera E.) cv. MRS2/5, plum rootstocks (Prunus cerasifera × P. salicina) × (P. cerasifera × P. persica) cv. Ishtarà and olive tree (Olea europaea L.) cv. Frantoio, cuttings were collected and treated with 0 mg.L-1 and 2500 mg.L-1 of indolebutyric acid (IBA) by immersion in the base of the cuttings. Exogenous genes can be introduced in plants by genetic transformation techniques. However, an efficient tissue culture system with high rates of plant recovery is necessary for gene introduction. This work aimed to define organogenesis and plant regeneration protocols for sweet orange varieties Natal, Valencia and Hamlin (Citrus sinensis L. Osbeck) and Rangpur lime (Citrus limonia L. Osbeck) which can be used in plant transformation experiments. Seeds of which teguments were removed, were germinated in vitro and maintained in the dark for three weeks, followed by one week at 16-h photoperiod (40 µmol m-2 s-1) and 27 ± 2°C.
